Background
Anthraxinfectionisinitiatedbytheinhalation,ingestion,orcutaneouscontactwithBacillusanthracisendospores.B.anthracisproducesthreepolypeptidesthatcomprisetheanthraxtoxin:protectiveantigen(PA),lethalfactor(LF),andedemafactor(EF).PAbindstotworelatedproteinsonthecellsurface;thesearetermedtumorepithelialMarker8(TEM8)/anthraxtoxinreceptor(ATR)andcapillarymorphogenesisprotein2(CMG2),althoughitisstillunclearwhichisphysiologicallyrelevant.FollowingPAbindingtoitsreceptor,PAiscleavedintotwofragmentsbyafurin-likeprotease.TheboundfragmentbindsbothLFandEF;theresultingcomplexisthenendocytosedwhichallowsthetranslocationofLFandEFintothecytoplasm.LFistheprimarytoxinofanthraxandfunctionsasahighlyspecificproteasethatcleavesmembersofthemitogen-activatedproteinkinasekinase(MAPKK)familyneartheiraminoterminus,interferingwithMAPKsignalingandinducingapoptosis.
