Background
Anti-ATG8antibodyisanUbiquitin-likeprotein(UBL)antibody.UBLproteinsandantibodiesfallintotwoclasses:thefirstclass,ubiquitin-likemodifiers(UBLs)functionasmodifiersinamanneranalogoustothatofubiquitin. ExamplesofUBLsareSUMO,Rub1(alsocalledNedd8),Apg8andApg12. Proteinsofthesecondclassincludeparkin,RAD23andDSK2,aredesignatedubiquitin-domainproteins(UDPs). Theseproteinscontaindomainsthatarerelatedtoubiquitinbutareotherwiseunrelatedtoeachother. IncontrasttoUBLs,UDPsarenotconjugatedtootherproteins. Apg8isrequiredforautophagy(intracellularbulkproteindegradation)inyeast. Starvedyeastcellstakeuptheirowncytoplasmintovacuolesthroughautophagicbodies. Autophagicbodiesformadouble-membranedstructurecalledtheautophagosome,whichsubsequentlyfuseswiththevacuole/lysosome.Thisprocesssimilarinmammals. Twosetsofgenes,APGandAUT,havebeenidentifiedwiththisprocess,andareresponsIBLefortwoubiquitin-likesystemsApg12andApg8,respectively. Apg12issynthesizedinitsmatureformandseemstohaveonetarget,Apg5. AlmostallApg12moleculesareconjugatedwithApg5. Aut2/Apg4processestheApg8/Aut7systematitscarboxy-terminalregion. Apg8existsintwoforms,oneismembraneboundthroughaphospholipid.Lipidation/activationofApg8ismediatedbyApg7andtransferredtoApg3andfinallyformsaconjugatewithphosphatidyl-ethanolamine(PE).Apg4cleavesApg8–PE,releasingApg8frommembrane. MorphologicalstudiesshowthatApg8localizesonthemembraneofintermediatestructuresoftheautophagosome;thistransientassociationseemstobeessentialforformationoftheautophagosome.BothApg12andApg8arehighlyconserved,withapparenthomologuesintheworm,mammalsandplants.Inhighereukaryotes,Apg8consistsofamultigenefamily.UsefulinCellBIOLOGy,MicrobiologyandSignalTransductionresearch.

ThispurifiedpolyclonalantibodyreactswithyeastAPG8bywesternblotandELISA.Althoughnottested,thisantibodyislikelyfunctionalinimmunohistochemistryandimmunoprecipitation. Thisantibodyusingthespecifiedconditionsmayrecognizeotherprominentintrinsicbands(UBLsortheirconjugates). OtherintrinsicbandsarereADIlydetectableinyeastlysatesatlowerantibodydilutions.Forimmunoblottinga14kDabandcorrespondingtoyeastAPG8isdetected. Mostyeastcelllysatescanbeusedasapositivecontrolwithoutinductionorstimulation.

Anti-ATG8antibodyisalsoknownasAnti-APG8andisanIgGfractionantibodypurifiedfrommonospecificantiserumbyamulti-stepprocesswhichincludesdelipidation,saltfractionationandionexchangechromatographyfollowedbyextensivedialysisagainstthebufferstatedabove.
