NF-kBwasoriginallyidentifiedasafactorthatbindstotheimmunoglobulinkappalightchainenhancerinBcells.Itwassubsequentlyfoundinnon-BcellsinaninactivecytoplasmicformconsistingofNF-kBboundtoIkB.NF-kBwasoriginallyidentifiedasaheterodimericDNAbindingproteincomplexconsistingofp65(RelA)andp50(NFKB1)subunits.Otheridentifiedsubunitsincludep52(NFKB2),cRel,andRelB.Thep65,cRel,andRelBsubunitsareresponsIBLefortransactivation.Thep50andp52subunitspossessDNAbindingactivitybutlimitedabilitytotransactivate.p52hasbeenreportedtoformtranscriptionallyactiveheterodimerswiththeNF-kBsubunitp65,similartop50/p65heterodimers.Lowlevelsofp52andp50homodimerscanalsoexistincells.Theheterodimersofp52/p65andp50/p65areregulatedbyphysicalinactivationinthecytoplasmbyIkB-alpha.IkB-alphabindstothep65subunitpreventingnuclearlocalizationandDNAbinding.ActivatorsmediatearapidphosphorylationofIkBbyIkBkinase(IKK)whichresultsinsubsequentubiquitinationandproteolyticdegradation.NF-kBisthentransportedtothenucleus,whereitactivatestranscriptionoftargetgenesthroughbindingtoNF-kBtargetsequenceswithinthepromoter.TheHTLV-IproteinTaxcaninduceconstitutiveNF-kBactivationthroughphosphorylationofbothIkB-alphaandIkB-beta.ThetransformingproteinTaxinhibitsp53transcriptionalactivitythroughtheNFkBsignalingpathway,specificallyviathep65(RelA)subunit.Theinhibitionofp53activityisdependentuponphosphorylationofp65(RelA)atS536bytheupstreamkinaseIKKß.Anti-NFkBantibodyisidealforCellBIOLOGy,NuclearSignaling,NeuroscienceandSignalTransductionResearch.
