Background
5-hydroxymethylcytosine(5-hmC)hasbeenrecentlydiscoveredinmammalianDNA.Thisresultsfromtheenzymaticconversionof5-methylcytosineinto5-hydroxymethylcytosinebytheTETfamilyofoxygenases.Sofar,the5-hmCbaseshavebeenidentifiedinPurkinjeneurons,ingranulecellsandembryonicstemcellswheretheyarepresentathighlevels(upto0,6%oftotalnucleotidesinPurkinjecells).Preliminaryresultsindicatethat5-hmCmayhaveimportantrolesdistinctfrom5-mC.Althoughitspreciserolehasstilltobeshown,earlyevidencesuggestsafewputativemechanismsthatcouldhavebigimplicationsinepigenetics:5-hydroxymethylcytosinemaywellrepresentanewpathwaytodemethylateDNAinvolvingarepairmechanismconverting5-hmCtocytosineand,assuchopenupentirelynewperspectivesinepigeneticstudies.Duetothestructuralsimilaritybetween5-mCand5-hmC,thesebasesareexperimentallyalmostindistinguishable.Recentarticlesdemonstratedthatthemostcommonapproaches(e.g.enzymaticapproaches,bisulfitesequencing)donotaccountfor5-hmC.Thedevelopmentoftheaffinity-basedtechnologiesappearstobethemostpowerfulwaytodifferentiallyandspecificallyenrich5-mCand5-hmCsequences.Anti-5hmCAntibodyisidealforresearchinEpigeneticsandGeneExpression.
