Background
Apoptosis,alsoknownasprogrammedcelldeath,playsmajorrolesindevelopmentandnormaltissueturnoverinadditiontotumorformation.Recentlyaproteinsimilartotheapoptosis-inducingfactor(AIF)wasclonedanddesignatedAIFL(alsoknownasAIFM3).AIFM3isexpressedubiquitouslyandispredominantlylocalizedtotheinnermembranesofmitochondria.UnlikeAIF,AIFM3doesnottranslocatetothenucleusuponinductionofapoptosis.However,overexpressionofAIFM3,likeAIF,inducedcytochromecreleasefromthemitochondria,cleavageofcaspase3,andultimatelyapoptosis,indicatingAIFM3inducesapoptosisthroughcaspaseactivation.MultipleisoformsofAIFM3areknowntoexist.
