AKT2isanisoformofthephosphoinositide-dependentserine-threonineproteinkinaseAKTandisenrichedininsulin-responsivetissuesandhasbeenimplicatedinthemetabolicactionsofthehormone. AKT2isaputativeoncogeneencodingaproteinbelongingtoasubfamilyofserine/threoninekinasescontainingSH2-like(Srchomology2-like)domains.FurThermore,AKT2wasshowntobeamplifiedandoverexpressedin2of8ovariancarcinomacelllinesand2of15primaryovariantumors.Over-expressionofAKT2contributestothemalignantphenotypeofasubsetofhumanductalpancreaticcancers.AKT2isageneralproteinkinasecapableofphophorylatingseveralknownproteins. AKT2mediatesmanyofthedownstreameventsofPI3-kinase(alipidkinaseactivatedbygrowthfactors,cytokinesandinsulin). PI3-kinaserecruitsAKT2tothemembrane,whereitisactivatedbyPDK1phosphorylation.Oncephosphorylated,AKT2dissociatesfromthemembraneandphosphorylatestargetsinthecytoplasmandthecellnucleus. AKT2hastwomainroles:(i)inhibitionofapoptosis;(ii)promotionofproliferation.

Althoughnottested,thisantibodyislikelyfunctionalinimmunohistochemistryandimmunoprecipitation.ThisproducthasbeenassayedbyimmunoblotagainstaHEK293celllysateandisreactiveata1:1,000dilutionshowingabandatapproximately60kDa. Aworkingdilutionof1:4,000to1:16,000issuggestedforthisproductinastandardcaptureELISAusingTMB(3,3",5,5"-Tetramethylbenizidine)code#TMBE-100asasubstratefor30minutesatroomtemperatureagainst0.1ugoftheimmunizingpeptide.Researchersshoulddetermineoptimaltitersforotherapplications.
Purity/Specifity
ThisaffinitypurifiedantibodyisdirectedagainsthumanAKT2.Theantibodydetectsbothunphosphorylatedandphosphorylatedformsoftheprotein.Theproductwasaffinitypurifiedfrommonospecificantiserumbyimmunoaffinitypurification. CrossreactivitywithAKT2fromotherspecieshasnotbeendetermined,however,thesequenceoftheimmunogenshows85%identitytomouseand92%identitywithrat,therefore,crossreactivityisexpected.
